Hi Eva,
I have started the modification f the C9 main vector (to put the tol2 transposon arms into it) but I have found out that the C9main stock, which we use for LR recombination actually is a mixture of two plasmids. One seems to be the right size (7.6 kb) but there is a smaller one around 5.5-6 kb. So is this normal (you need the second plasmid to be able to grow the C9main in the ccdB, cells, e.g normally is co-isolated (co-transformed) with C9main form (into) these cells) or the vector is contaminated.

Regarding the screen it looks quite bad at the moment, it seems that there is a general contamination of the plasmids because also 0102 which was cloned quite early shows it. Maybe already the stock was contaminated. We will check now each plasmid made by restriction digest, because the second one is only visible after digestion. With undigested plasmid it is hard to see. If we will find thta the ratios between different preps are different the biological part of the project is actually dead. Let's hope they will be at least comparable.

I am just sorting the plasmids for the big digest :)
I have digested 19 in a test and just 1 was contaminated (0410). lets hope the problem is not as big
